چکیده

The corpus callosum is the brain’s primary pathway for interhemispheric communication between the left and right hemispheres, and abnormal function and anatomy of the corpus callosum have been associated with a variety of disorders in humans and animals. In this study, we mapped quantitative trait loci (QTLs) modulating corpus callosum size in mice. By using mouse brain sections and stereological techniques, we estimated the midsagittal corpus callosum size in 191 BXD recombinant inbred mice and their parent strains (C57BL/6J and DBA/2J). Analysis showed midsagittal corpus callosum size to be a heritable trait (h = .42). We detected a suggestive QTL modulating corpus callosum size near the marker S10Gnf071.990 on chromosome 10 at approximately 73 Mb, in close proximity to a previously defined QTL modulating striatal volume. We also found correlations between corpus callosum size and other behavioral and anatomical phenotypes. Overall, our results implicate a genetic role in modulation of corpus callosum size.

Localization of two new X-linked quantitative trait loci controlling corpus callosum size in the mouse.

Corpus callosum (CC) size is a complex trait, characterized by a gradation of values within a normal range, as well as abnormalities that include a small or totally absent CC. Among inbred mouse strains with defects of the CC, BTBR T(+)tf/J (BTBR) mice have the most extreme phenotype; all animals show total absence of the CC and severe reduction of the hippocampal commissure (HC). In contrast, ...

Quantitative Trait Loci for Interhemispheric Commissure Development and Social Behaviors in the BTBR T+ tf/J Mouse Model of Autism

BACKGROUND Autism and Agenesis of the Corpus Callosum (AgCC) are interrelated behavioral and anatomic phenotypes whose genetic etiologies are incompletely understood. We used the BTBR T⁺ tf/J (BTBR) strain, exhibiting fully penetrant AgCC, a diminished hippocampal commissure, and abnormal behaviors that may have face validity to autism, to study the genetic basis of these disorders. METHODS W...

Tests of genetic allelism between four inbred mouse strains with absent corpus callosum.

Inbred mouse strains that lack the corpus callosum connecting the cerebral hemispheres in the adult differ from the C57BL/6J strain at several relevant but unknown loci. To identify at least one major locus that influences axon guidance, different strains showing phenotypically similar defects were crossed to test for allelism. If the F1 hybrid between two strains with the same brain defect is ...

Genetic control of the mouse cerebellum: identification of quantitative trait loci modulating size and architecture.

To discover genes influencing cerebellum development, we conducted a complex trait analysis of variation in the size of the adult mouse cerebellum. We analyzed two sets of recombinant inbred BXD strains and an F2 intercross of the common inbred strains, C57BL/6J and DBA/2J. We measured cerebellar size as the weight or volume of fixed or histologically processed tissue. Among BXD recombinant inb...

Identifying Human Disease Genes through Cross-Species Gene Mapping of Evolutionary Conserved Processes

BACKGROUND Understanding complex networks that modulate development in humans is hampered by genetic and phenotypic heterogeneity within and between populations. Here we present a method that exploits natural variation in highly diverse mouse genetic reference panels in which genetic and environmental factors can be tightly controlled. The aim of our study is to test a cross-species genetic map...
